Glial cell-derived neurotrophic factor promotes the proliferation and differentiation of midbrain-derived neural stem cells into dopaminergic neurons

To explore the ability of glial cell line-derived neurotrophic factor (GDNF) to proliferate and differentiate mesencephalic neural stem cells (mNSCs) and express pituitary homeobox family factor 3 (Pitx3), orphan nuclear receptor-related factor 1 (Nurr1) and tyrosine The effect of amino acid hydroxylase (TH) protein. Methods: The primary mNSCs were cultured in vitro on E14.5 d rat midbrain tissues, and the differentiation experiment was carried out after three generations of proliferation culture; immunofluorescence method was used to detect the expression of Pitx3, Nurr1 and TH proteins in cultured and differentiated mNSCs. Results: Under the action of GDNF, the diameter and number of mNSCs cloned in vitro increased, and the expression of Pitx3, Nurr1 and TH protein in differentiated neurons increased (P <0.05). Conclusion: In vitro culture conditions, GDNF can promote the proliferation of mNSCs and differentiation into dopaminergic neurons.
